A neuropeptide encoded by the KISS1 gene that acts as the master upstream regulator of the reproductive hormone axis. Studies have investigated its role in stimulating GnRH release and modulating reproductive signaling pathways.
Kisspeptin is a family of peptides (kisspeptin-54, -14, -13, -10) derived from proteolytic cleavage of the KISS1 gene product. All forms share a common C-terminal decapeptide sequence that binds to and activates the kisspeptin receptor (KISS1R/GPR54) on GnRH neurons in the hypothalamus. This activation is the primary physiological trigger for pulsatile GnRH secretion, which in turn drives the pituitary release of luteinizing hormone (LH) and follicle-stimulating hormone (FSH). Kisspeptin integrates metabolic, stress, and photoperiod signals to coordinate reproductive function. Loss-of-function mutations in KISS1 or KISS1R cause hypogonadotropic hypogonadism.
